  • Abstract:

    [Objective] To study the mitochondrial proteome of mouse brain at different developmental stages by mass spectrometry in order to provide data support for the study of nervous system diseases. [Methods] The brain cells of mice at 15 days after embryo, 8 days after birth and 60 days after birth were selected from the Laboratory Animal Center of China Medical University. The lysate was prepared, and the lysate was mixed with the mouse brain cell samples in a ratio of 3:1. After a series of routine operations, the brain mitochondrial proteins were extracted. The database was retrieved by using the analysis software PDQuest, and the protein types were identified by mass spectrometry. [Results] The protein expression profiles were obtained, including phosphoprotein P19, actin, αenolase, actin 41[fragment], 28×103 hot acid stabilized phosphoprotein, 40S ribosomal protein S3a, 3-mercaptopyruvate sulfur transferase and C140RFL66 homologous protein. Ten protein spots were selected. According to the comparison results, the protein pairing ratio of mass spectrometry was the highest 96% and the lowest 75%, which was higher than the lowest protein pairing ratio of 68%-72% in the group, showing accurate pairing effect and short pairing time. [Conclusion] The maximum ratio of this method is 96%, and the shortest ratio time is 38ms, which lays a foundation for the study of cell injury.
